MetaCyc Enzyme: γ-F420-2:α-L-glutamate ligase

Gene: cofF Accession Numbers: G-9428 (MetaCyc), MJ1001

Species: Methanocaldococcus jannaschii

Summary:
The cofF (MJ1001) gene of Methanocaldococcus jannaschii, a homolog of the Escherichia coli K-12 rimK gene, was cloned and expressed in Escherichia coli. The resulting monomeric protein was shown to be a γ-F420-2:α-l-glutamate ligase, the enzyme that caps the γ-glutamyl tail of the hydride carrier coenzyme F420 [Li03].

Molecular Weight of Polypeptide: 33.112 kD (from nucleotide sequence), 37 kD (experimental) [Li03 ]

Gene-Reaction Schematic

Enzymatic reaction of: γ-F420-2:α-L-glutamate ligase

EC Number: 6.3.2.32

oxidized coenzyme F420-2 + L-glutamate + ATP <=> coenzyme α-F420-3 + ADP + phosphate + H+

The reaction direction shown, that is, A + B ↔ C + D versus C + D ↔ A + B, is in accordance with the direction in which it was curated.

The reaction is physiologically favored in the direction shown.

Summary:
Vmax was 0.72 nmol/min/mg protein [Li03].

Activators (Allosteric): Mn2+ [Li03] , Mg2+ [Li03]
