Summary:
A novel alliinase was purified to homogeneity from roots of onion (Allium cepa). The purification yielded two isoforms with similar substrate preference. However, they differed from each other in their glycosylation with isoform II having additional terminal mannose structures. A cDNA isolated from the onion root showed identical amino acid sequence to isoform II confirming this isoform as the allinase residing in the root. Unlike other alliinases the protein was shown to have both cysteine lyase as well as cysteine sulfoxide lyase (allinase) activities. The protein catalyzed the conversion of isoallin to 1-propenylsulfenate with high efficiency. The protein had isoelectric points of 7.6, 7.9, 8.1 and 8.3. Vmax for isoallin as substrate was 21,524 nkat/mg. The alliinase was mainly expressed in onion roots [Lancaster00].
