Summary:
The Arabidopsis thaliana gene ITPK1 encodes a protein with myo-inositol-1,3,4-trisphosphate 5/6-kinase activity. The gene was recombinantly expressed in E.coli and purified to test its substrate specificity. The enzyme was shown to use D-myo-inositol (1,3,4)-trisphosphate as a substrate to produce both D-myo-inositol (1,3,4,5)-tetrakisphosphate and D-myo-inositol (1,3,4,6)-tetrakisphosphate in a 3:1 ratio [Wilson97]. No evidence of the phosphorylation of D-myo-inositol (1,4,5)-trisphosphate, Ins(1,4)P2, Ins(1)P, Ins(2)P, Ins(3)P or cyclic Ins(1,2)P by the recombinant protein was found [Wilson97].
