MetaCyc Enzyme: 4-oxalocrotonate decarboxylase

Gene: amnE Accession Number: G-11252 (MetaCyc)

Species: Pseudomonas sp. AP3

Subunit composition of 4-oxalocrotonate decarboxylase = [AmnE]
         4-oxalocrotonate decarboxylase monomer = AmnE

Summary:
4-oxalocrotonate decarboxylase has been purified from Pseudomonas sp. AP3 to homogeneity [Takenaka00]. The protein was found to form a large complex (300 kDa) with the hydratase encoded by amnF, which catalyzes the next step of the pathway. The ratio of decarboxylase to hydratase subunits within the complex was 1:1.

The enzyme required either Mg2+ or Mn2+ for activity, and was sensitive to several metal ions, including Fe2+, Fe3+, Ag+ and Hg2+ [Takenaka00].

Molecular Weight of Polypeptide: 27.456 kD (from nucleotide sequence), 29.0 kD (experimental) [Takenaka00 ]

Enzymatic reaction of: 4-oxalocrotonate decarboxylase

EC Number: 4.1.1.77

(3E)-2-oxohex-3-enedioate + H+ <=> 2-oxopent-4-enoate + CO2

The reaction direction shown, that is, A + B ↔ C + D versus C + D ↔ A + B, is in accordance with the Enzyme Commission system.

The reaction is favored in the direction shown.

In Pathways: 2-aminophenol degradation

Cofactors or Prosthetic Groups: Mg2+ [Takenaka00], Mn2+ [Takenaka00]

Inhibitors (Unknown Mechanism): Fe2+ [Takenaka00] , Fe3+ [Takenaka00] , Ag+ [Takenaka00] , Cu2+ [Takenaka00]

pH(opt): 7.5 [Takenaka00]
