MetaCyc Enzyme: dihydrofolate synthetase

Summary:
The Arabidopsis genome was searched for sequences similar to dihydrofolate synthetase/folylpolyglutamate synthetase (DHFS/FPGS) signatures and four cDNA's were retrieved. The molecular functions of the cDNA's were tested by functional complementing mutant yeasts defect in either DHFS or FPGS activity. Protein extracts from the complemented yeast were used to confirm the specific function of the enzyme in vitro. One of the cDNA's, named AtDFA (At5g41480), encoded for the dihydrofolate synthetase (DHFS) in Arabidopsis thaliana catalyzing the conversion from 7,8-dihydropteroate to 7,8-dihydrofolate but did not exhibit any folypolylglutamate synthase activity [Ravanel01]. The mitochondrial location of the enzymes marks this compartment as the only one with a complete set of all enzymes involved in folate biosynthesis.
