Arabidopsis has two functional genes encoding proline dehydrogenase - POX1 and POX2 [Funck10]. The two proteins have non-redundant, although partially overlapping physiological functions. Proline dehydrogenase 1 appears to be the dominant isoform under most conditions and in most tissues, while proline dehydrogenase 2 is specifically upregulated during salt stress (when POX1 is repressed) [Funck10].

proline dehydrogenase 2 is expressed in the vascular tissue and in the abscission zone of petals, sepals, stamina, pistils and siliques. It is Down-regulated by high succrose and up-regulated by L-proline and drought. When expressed in yeast the protein rescued the Pro metabolism defect of a Δput1 mutant strain [Funck10].
