Summary:
RatA is a toxic protein that inhibits initiation of translation by specifically associating with the free 50S subunit of the ribosome and inhibiting 70S ribosome formation. RatA is unable to dissociate 70S ribosomes and has no effect on cellular mRNAs [Zhang11a].

Inducing expression of RatA causes inhibition of cell growth [Brown03, Zhang11a]. Deletion of the orthologous pasTI locus alone in the uropathogenic E. coli CFT073 causes a defect in colonizing the kidneys of a mouse UTI model system and a defect in persister cell formation. In contrast, deletion of ratAB does not affect persister cell formation in E. coli K-12 MG1655 and does not increase sensitivity to nitrosative stress caused by acidified sodium nitrite (ASN). However, low-level expression of RatA does increase resistance to ASN [Norton12].
