Summary:
ErpA is an A-type iron-sulfur protein; it can bind both [2Fe-2S] and [4Fe-4S] clusters. In vitro, ErpA can transfer a [4Fe-4S] cluster to apo-IspG [Loiseau07].

Functional redundancy between the three A-type carriers (ATCs) in E. coli, ErpA (an ATC-I) and IscA and SufA (both belonging to the ATC-II family), has been investigated [Vinella09].

erpA is essential for aerobic growth and under conditions of anaerobic respiration; the conserved cysteine residues are important for in vivo function. The growth defect of an erpA null mutant can be partially rescued by expression of the eukaryotic MVA-dependent pathway for IPP synthesis (mevalonate pathway I), whose enzymes do not contain Fe-S clusters. An erpA mutant contains 5-6-fold less quinone than wild type [Loiseau07]. An erpA null mutant contains no active Fdh-N, Fdh-O, and nitrate reductase [Pinske12] and no detectable hydrogenase 1 or hydrogenase 2 activity [Pinske12a] due to the absence of the [Fe-S] cluster-containing small subunits of these enzymes.

Transcription of erpA is under the direct control of IscR [Giel06].
