Summary:
Isolation of spontaneous N4 bacteriophage-resistant mutants of E. coli K-12 strain SC1100, cloning of the affected genes, and subsequent complementation experiments revealed that nfrA is important in N4 adsorption. The 96 kDa nfrA gene product was found to be associated with the outer membrane in SDS-PAGE of outer-membrane fractions, though maxicell analysis found a small amount of NfrA associated with the inner membrane along with its 98 kDa precursor. This may be due to inefficient processing and transport of NfrA in maxicells. A mutation in the nfrB gene, directly upstream of nfrA and also important for N4 adsorption, did not affect NfrA transport to the outer membrane [Kiino89].
